As the focus shifts towards enhancing the quality of life and improving cancer prevention and early intervention strategies, there is a growing interest in exploring alternative therapies, including herbal medicine.

One such herbal remedy with potential anti-cancer properties is Atractylodes macrocephala Koidz (AMK), a traditional East Asian medicine used for centuries to address various ailments, including cancers. These compounds, including GLY, LPG, ASI, D-Serin, and others, exhibited interactions with 260 target genes associated with diverse cellular pathways.
 
Furthermore, the study revealed 37 AMK phytochemicals linked to gastrointestinal (GI) cancer, highlighting the herb's specific relevance to gastric cancer treatment. Notably, compounds like atractylenolide I, atractylenolide III, and palmitic acid showed direct associations with critical gastric cancer-related genes such as TNF, IL6, BCL2, PTEN, CTNNB1, and SRC.
 
Experimental Insights into AMK's Anti-Cancer Mechanisms In Vitro Experiments
-Cytotoxicity and Cell Cycle Arrest: AMK extract demonstrated concentration-dependent cytotoxic effects on AGS human gastric aden ocarcinoma cells. Notably, it induced G1 phase cell cycle arrest, a characteristic feature of anti-cancer agents.
 
-Apoptosis Induction: AMK extract triggered apoptosis in AGS cells through mitochondrial pathway activation, as evidenced by increased ROS production, alterations in mitochondrial membrane potential, and activation of caspases involved in intrinsic apoptotic pathways.
 
-Modulation of Signaling Pathways: AMK extract influenced key signaling pathways such as MAPK (mitogen-activated protein kinase), leading to reduced cell proliferation and migration capabilities of AGS cells.
 
-Inhibition of Tumor Growth: In a 3D spheroid model mimicking tumor growth, AMK extract significantly suppressed spheroid formation, further validating its anti-cancer potential.
